The hyposmotic test is an additional test in the study of male fertility, as it allows a distinction to be made between sperm with a dysfunctional membrane and sperm without alterations in the membrane. This means that, thanks to the addition of a hypotonic solution to the semen, it is possible to differentiate between dead and living spermatozoa.
This difference is due to the fact that the living spermatozoa have their membrane intact and the solution will pass into the spermatozoa, causing the tail to roll up. However, the membrane of the dead sperm is altered and the hypotonic solution is not introduced, so their tail will be smooth.
